Development of monolayer lipid nanoparticles targeting apolipoproteins B100 and B48

About

This is a research project proposal called ApoLipoNano focused on developing innovative lipid nanoparticles to fight cardiovascular disease and hyperlipidemia.

The core idea is to create functionalized solid lipid nanoparticles (SLN-ApoB) that directly target apolipoproteins B100 and B48 — the proteins responsible for transporting harmful LDL, VLDL, and chylomicron particles in the bloodstream. Rather than blocking cholesterol synthesis like statins or modulating receptors like PCSK9 inhibitors, this approach aims to neutralize and clear the atherogenic carriers themselves.

The project runs over 36 months across five phases: nanoparticle design, physicochemical characterization, in vitro cell testing, in vivo preclinical animal studies, and regulatory filing. The primary goal is a 30% reduction in plasma LDL in preclinical models, with a safety profile suitable for Phase I clinical trials.

The total budget is €850,000, with €600,000 requested in funding, involving a multidisciplinary team of researchers, PhD students, and postdocs. If successful, the technology could serve as a new therapeutic class for patients intolerant to existing treatments, targeting a global market estimated at over €15 billion.
