A First-in-Class Precision Oncology Platform: RAGE-Targeted ADC Therapy for Colorectal and Broader Solid Tumour Indications.

Viamab Therapeutics is pursuing accelerated clinical development of the world's first RAGE-targeted antibody–drug conjugate, targeting first-in-class trial entry in colorectal cancer as our lead indication, with broader pan-cancer potential across RAGE-expressing tumour types.

Our mission is to bring genuinely novel treatment options to patients with limited or no effective alternatives — where the science is compelling and the unmet need is greatest.

1.9 million people are diagnosed with colorectal cancer globally each year — rising to 3.2 million by 2040. Just under half will develop advanced disease, where 5-year survival falls below 20%, existing third-line options deliver response rates of just 1–7%, and ~95% of patients have no ADC options. This represents a profound unmet need for effective targeted treatment.RAGE (Receptor for Advanced Glycation End products) expression increases progressively with colorectal cancer stage, with low expression observed in early-stage tumours (19% in stage II disease) and substantially higher expression in advanced stages, reaching 81% in stage III and 100% in stage IV tumours.

No ADC currently targets RAGE in any tumour type. RAGE is also overexpressed across pancreatic, breast, ovarian, prostate, gastric, renal and other solid tumour cancers — all indications where advanced-stage patients have limited or no targeted treatment options.

Current traction (preclinical):

• First-on-target RAGE ADC with validated receptor-mediated internalisation and payload delivery.

• In vitro efficacy demonstrated in CRC models.

• Supporting data across ovarian, prostate and triple-negative breast cancer (xenograft, syngeneic/orthotopic).

• Mouse toxicology complete — clean profile to date

• Strong IP: one granted patent, one pending.

• Key advisor credited on two FDA-approved ADCs — KADCYLA and ELAHERE

Investment opportunity: $30M total to Phase 1 readout comprising of $15M to IND-ready and an additional $15M through Phase 1 — leveraging Australian clinical trial environment incentives to maximise capital efficiency. The raise provides natural inflection points throughout the timeline using key development milestones that help de-risk the investment.

Beyond investment, we are actively exploring licensing and co-development partnerships and open dialogues with pharma and biotech groups who see potential strategic value in a first-on-target RAGE ADC.
