HεκKI Human IgE Knock-In Mice — Live Animal Model for Anti-IgE Drug Discovery

Croyez Bioscience (Taiwan) offers the HεκKI Platform — Human IgE (ε, κ) Knock-In Mice — a live animal model built for investigating human IgE inhibition, the central mediator of type I hypersensitivity reactions (allergic asthma, allergic rhinitis, atopic dermatitis).

How it works: the transgenic mouse genome has its Cγ1 constant region replaced by the human immunoglobulin Cε constant region, and its Cκ constant region replaced by the human immunoglobulin Cκ constant region. After immunization, HεκKI mice produce humanized IgE-secreting B cells and antigen-specific chimeric human IgE, enabling in vivo human IgE inhibition studies with antibodies, small molecules, or vaccines.

Key features:

30-fold higher human IgE response than mouse IgE

Does not produce mouse IgG1, so the humanized signal is clean and specific

Validated in published literature (Lu CS, et al. Allergy. 2015) and in a real anti-IgE drug case study: a customer developing a treatment for chronic spontaneous urticaria (CSU) used HεκKI mice to show their candidate (UB-221) reduced serum free-chimeric-IgE by more than 90% (Kuo BS, et al. J Clin Invest. 2022)

This complements our antibody and antigen product lines (allergen-specific IgE and antigen library) with a live in vivo model. The animal model can be imported by qualified customers for their own contract research or drug development programs, spanning diagnostics, infectious disease, vaccines, and biologics.
