Rapid access to a suitable production strain is a key determinant of success in recombinant protein production (RPP) using Pichia pastoris (reclassified as Komagataella phaffii). This course provides a comprehensive introduction to host strain engineering, that combines a solid theoretical foundation with extensive hands-on training in modern molecular biotechnology techniques.

Participants will gain an overview of the genetic toolbox available for Pichia, covering established and emerging strategies for host strain development and recombinant protein production. The course addresses the principles of rational strain engineering, expression cassette design, genome editing, and experimental workflows required to generate highly productive recombinant strains. Practical laboratory sessions complement the lectures by providing direct experience with state-of-the-art methodologies and experimental procedures routinely applied in Pichia engineering.

A particular emphasis is placed on advanced genome engineering technologies, with dedicated sessions covering the application of different CRISPR–Cas systems for precise and efficient genetic modification. Participants will learn both the underlying concepts and their practical implementation in Pichia strain engineering workflows.

This course will take place entirely at the Institute of Molecular Biotechnology, TU Graz, providing participants with access to modern research facilities and expert supervision throughout the practical training.

Dedicated discussion sessions with experts in the field will provide opportunity to address methodological questions and discuss current challenges as well as future developments in Pichia research and recombinant protein production.

As in the years before, the interactive Hands-on-Pichia course offers a great opportunity for graduate students, researchers, and professionals to expand their expertise in Pichia host strain engineering while gaining practical experience with cutting-edge molecular biotechnology techniques.
