Development of monoclonal antibodies to prevent and treat infections from Flaviviruses (HORIZON-HLTH-2027-01-DISEASE-06)

Project manager _ Pre-award at Fundación Pública Miguel Servet - Navarrabiomed

Pamplona, Spain

About

The Protein Crystallography and Structural Immunology Unit at Navarrabiomed (Spain) is interested in participating in European consortia developing monoclonal antibody-based therapies against emerging and re-emerging flaviviral infections.

We investigate antibody-antigen recognition using recombinant expression in E. coli, Sf9 and CHO systems, ÄKTA-based purification of antigens and antibody/Fab constructs, and BLI/SPR measurements of affinity and binding kinetics. Gryphon-assisted crystallisation, Zeiss V201 crystal inspection and routine access to the ALBA synchrotron support X-ray analysis of selected antigen-Fab complexes.

We could contribute to the detailed structural characterisation of existing monoclonal antibody candidates against eligible flaviviruses, including the identification of recognised epitopes, the investigation of antibody-antigen interaction interfaces and the analysis of molecular determinants of specificity. Structural and computational approaches could also support the identification of conserved antigenic regions, assessment of the potential impact of viral sequence variation and rational optimisation of antibody candidates.

For flaviviruses where antibody-dependent enhancement (ADE) is a relevant concern, our structural studies could help interpret antibody-binding patterns and guide the design of complementary functional assessments by specialised virology partners.

Our previous experience in structural vaccinology, antigen engineering and antibody-antigen recognition provides a foundation for supporting therapeutic antibody development.
